The incorporation of C14-orotic acid into nucleic acid pyrimidines in vitro.

چکیده
The incorporation in vivo of compounds labeled with N15 or Cl4 into the purines and pyrimidines of nucleic acid has been repeatedly demonstrated (e.g. (l-3)). W e wish to report observations on the incorporation of radioactive erotic acid in vitro into the pyrimidines of the nucleic acid of rat liver slices. It is believed that this procedure will readily permit the study of precursors of nucleic acid and other phases of nucleic acid metabolism. It may possibly provide an objective approach to the study of nucleic acid metabolism in tumors.

Studies on fluorinated pyrimidines. V. Effects on nucleic acid metabolism in vitro.
To gain further information on the mechanisms by which the fluorinated pyrimidines and their nucleosides inhibit various processes of nucleic acid biosynthesis, studies similar to those re ported in the preceding paper (6) were carried out in vitro in glycolyzing suspensions of the Ehrlich ascites cells system used by LePage for studies of purine biosynthesis (20). In this work the following la...

A revival of interest has taken place in recent years in the possibility of interference with mam malian nucleic acid biosynthesis by means of an tagonists of the free pyrimidines. Early studies appeared to show that, while orotic acid (1, 48) and the sugar derivatives of the pyrimidines (19, 20) are extensively incorporated into both RNA and DNA, the incorporations of free uracil, cytosine, an...
متن کاملThe incorporation of radioactive orotic acid into the nucleic acid pyrimidines of animal and human tumors.
The incorporation of radioactive orotic acid in vitro into the pynimidines of the nucleic acid of animal tissues has been studied (6, 7). In this paper data are presented on the incorporation of radioactivity into the pynimidines of the nucleic acids of animal and human tumors. It is impos sible to make any more than tentative conclusions on the differences of tumor tissue from the “non mal t...
متن کاملA survey of the metabolism of orotic acid in the rat.
Orotic acid has been demonstrated to be an effective precursor of the nucleic acid pyrimidines in animal tissues, both in viva (1, 2) and in vitro (3,4). It is a growth-stimulating factor for certain pyrimidineless mutants of microorganisms (5-8) and may be partially replaced by oxalacetic acid (9) or ureidosuccinic acid (10, 11) in some strains. The possibility that erotic acid, and, hence, th...
متن کاملIn vitro antimalarial activity of nucleic acid precursor analogues in the simian malaria Plasmodium knowlesi.
Analogues of nucleic acid precursors were screened for antimalarial activity in Plasmodium knowlesi by using an in vitro culture system. Activity was assessed by the degree of inhibition of incorporation of l-[methyl-(14)C]methionine into protein and of [8-(14)C]adenosine and [6-(14)C]orotic acid into ribonucleic acid and deoxyribonucleic acid. The incorporation of adenosine or orotic acid was ...
